Auraptene (Aur) is a simple coumarin found in the peels of citrus fruits such as lemon, grapefruit, and orange. We examined the latent mechanism of Aur and its protective role in alcohol-induced hepatic and intestinal damage by both in vitro and in vivo experiments. In vivo model of acute alcoholic hepatosteatosis were established by intragastrically administered ethanol to male C57BL/6 mice. In vitro, HCT116 cells were treated with ethanol. AML12 cells were challenged with supernatant from HCT116 cells treated with ethanol. In vivo, we identified that Aur treatment could reduce serum levels of ALT and AST, as well as the decrease lipid accumulation and suppress inflammation in liver. In the colon, Aur increased the expression of SIRT1 and phosphorylated AMPKα, while inhibiting the expression of P2x7R and IL-1β. In addition, Aur up-regulated the expression of Nrf2 in the liver and colon, and it alleviated the significant intestinal barrier damage induced by ethanol consumption. In vitro, it was detected the release of mediators from colon cells might contribute to Aur's improvement of hepatic injury in AML12 cells. These findings further explored the mechanism of Aur to improve acute alcohol exposure induced intestinal liver injury, and opened a new therapeutic window.
